Procedure:
1. Prepare a thin smear on clean dry glass slide.

6. Rinse with water. Blot dry.

Principle:
The granules stain purple-black against the light green counterstained cytoplasm. This helps to distinguish Corynebacterium diphtheria from most of the short nonpathogenic diphtheroides which lack granules.

Intended Use: Staining Solution for Microbes

Principle:
This differential staining technique is useful for identification of the tubercle bacillus, other Mycobacteria and Nocardia, which depends on the chemical composition of the bacterial cell wall. Because of the difficulty in staining these organisms with ordinary dyes, basic dyes in the presence of controlled amounts of acid are used. Generally, heat must be applied during the staining procedure, or wetting agents must be used, to aid dye penetration. Organisms exhibiting the property of acid-fastness, once stained, are not easily decolourized by alcohol.

PROCEDURE
1. Prepare a smear on a clear, dry glass slide.
2. Allow it to air dry and fix with gentle heat.
3. Flood the smear with Carbol Fuchsin stain. Heat to steaming for 5 minutes with a low flame.
Do not boil the stain and do not permit drying of the smear.
4. Allow it to stand for 5 minutes without further heating.
5. Wash in the running tap water.
6. Decolourize with Acid Fast Decolourizer for 2 minutes or until no more stain comes off in the
washings. (If washing is not thorough, may get false positive results).
7. Wash with the tap water.
8. Counterstain for 30 seconds with Methylene Blue.
9. Wash with tap water. Dry in air. Examine under an oil immersion objective.

RESULTS
1.Acid fast organisms: Bright red
2 All other organisms: Blue

PROCEDURE
1.Prepare a smear on a clear, dry glass slide.

2.Allow it to air dry and fix with gentle heat.

3.Flood the slide with Crystal Violet.
4.Allow the stain to be in contact with the smear for 1-2 minutes.

5.Wash in slow-running water, just enough to remove excess of dye.
6.Flood the smear with Iodine, drain and flood again with Iodine for 1 minute.

7.Wash with decolourizer (alcohol) for about 5-15 seconds. Wash the slide to stop the action of decolorize.

8.Flood with safranin for 1 minute, wash very lightly.

9.Blot dry and examine under oil immersion objective.

RESULTS

Gram-positive organisms: Violet colored.
Gram-negative organisms: Pinkish red colored.

NEISSER′S METHYLENE BLUE

 NEISSER′S METHYLENE BLUE

Staining Solution for Metachromatic staining.

Principle:
Well-developed granules of volutin (polyphosphate) may be seen in unstained wet preparations as round refractile bodies within the bacterial cytoplasm. With basic dye, they tend to stain more strongly than the rest of the bacterium, and with toluidine blue or methylene blue they stain metachromatically, a reddish purple colour. They are demonstrated most clearly by special methods, such as Albert’s and Neisser’s, which stain them dark purple but the remainder of the bacterium with a contrasting counterstain. The diphtheria bacillus gives its characteristic volutin-staining reactions best in a young culture (18-24 hours) on a blood or serum medium.

6. Pour Neutral Red solution, keep it for 3 min.
7. Wash with distilled water.
8. Allow the smear to air dry.

9.Examine under oil immersion microscope.

PROCEDURE:
1. Bring the section to slide using suitable adhering agent.
2.Slightly warm to melt the wax.
3.Remove the wax with Xylene by giving 3 times washing.
4. Remove the Xylene in 3 times washing with Dehydrant.
5. Dip the smear into distilled water for 3 minutes.

6. Dip the smear in Heamatoxylin (Harris) stain for 1min.
7. Wash with Distilled water.
8. Dip the smear in Eosin stain for 60 seconds.
9. Wash with distilled water for 15 seconds.
10. Rinse with Dehydrant and Xylene till it gets cleared.
11. Mount with glass cover using a drop of DPX.

HEMATOXYLIN & EOSIN STAIN

 Intended use:

HISTOPATHOLOGY STAIN.

Principle:
Alum acts as mordant and hematoxylin containing alum stains the nucleus light blue. This turns red in presence of acid, as differentiation is achieved by treating the tissue with acid solution. Bluing step converts the initial soluble red color within the nucleus to an insoluble blue color. The counterstaining is done by using eosin which imparts pink color to the cytoplasm.
